정제 함량 균일성 실패

정제 함량 균일성 실패

Tablet content uniformity describes the consistency of active pharmaceutical ingredient, or API, content between individual tablets. Failures can originate during initial API distribution, segregation after blending, or ingredient redistribution during wet granulation and drying. The investigation should identify where composition changes before selecting a corrective action. For production and process engineering teams, the most useful question is where a previously acceptable material becomes nonuniform. This guide follows the material from premixing through drying, transfer, and compression, with practical checks for each stage.

Why Consistent Tablet Weight Does Not Prove Uniform API Content

Tablet mass and API concentration both influence the amount of API in an individual tablet. If concentration varies through a blend, tablets of similar weight can contain different doses. A composite batch assay can also conceal variation between individual units because it combines material before measurement.

For illustration, two tablets may each weigh 200 mg. At API concentrations of 4% and 6%, they contain 8 mg and 12 mg of API, respectively. This arithmetic example explains why weight checks alone cannot diagnose concentration differences; it does not represent a product specification or acceptance limit.

Use the applicable pharmacopoeial procedure and approved product specification to assess dosage uniformity. Weight variation is an allowable dosage uniformity approach only under the relevant conditions; routine tablet weight checks should not be treated as a universal substitute for content uniformity testing. [1]

For broader compression and appearance problems, see Jianpai’s overview of common tablet manufacturing defects.

Identify the Pattern Before Changing the Process

Compare individual results with production time, material transfers, hopper refills, and process interruptions. Patterns can guide an investigation, although none proves a root cause on its own.

Observed pattern Possible explanation to investigate Useful evidence
Similar tablet weights with variable API results Concentration differences rather than mass variation Individual tablet results, corresponding weights, blend data
Acceptable blend samples followed by variable tablets Segregation after sampling or unrepresentative blend samples Transfer records, sampling method, tablets across the run
API results drift as compression progresses Changing material composition at the feeder Beginning, middle, and end samples; hopper refill timing
Different assays across granule size fractions Migration, nonuniform granulation, or selective breakage Fraction assays, size distribution, collected fines

Inadequate API Distribution During Premixing

Investigate Low Dose Ingredients and Agglomerates

A low API loading makes local distribution especially important. If the API remains in agglomerates, an apparently well-mixed bulk powder may still contain small regions with different concentrations. A small number of relatively large API particles can also limit the consistency of distribution at the individual dose scale.

Review incoming API particle size, agglomeration, dispensing, charging order, and premix preparation. Assess whether deagglomeration, a suitable carrier, preblending, or geometric dilution is appropriate for the formulation. Finer particles can improve dispersion in some systems, but excessive size reduction can increase cohesion and handling difficulties.

Establish a Supported Blending Endpoint

Blending performance depends on equipment geometry, fill level, speed, material properties, and charging sequence. Extending the mixing time without evidence may fail to break persistent agglomerates or address segregation during discharge.

Use development studies to establish an operating range and sampling strategy appropriate to the formulation. Review sample location, sample mass, and analytical capability together. A large sample may average out concentration differences that matter at tablet scale.

에이 pharmaceutical powder blender should be assessed using representative material and its intended loading range. The trial should demonstrate API distribution rather than relying only on mixing time or a visual inspection.

Segregation After an Acceptable Blend

Follow the Material Through Discharge and Transfer

Differences in particle size, density, and shape can promote separation during discharge, free fall, vibration, conveying, or hopper flow. A satisfactory sample collected inside a blender does not establish that material arriving at the tablet press remains uniform.

Map the actual transfer route, including intermediate containers and holding steps. Examine drop heights, discharge restrictions, vibration, hopper geometry, and pneumatic conveying conditions where applicable. Compare samples before and after suspected transfer steps using an appropriate sampling protocol.

When results change following a hopper refill or near the end of a run, investigate changes in feed composition alongside press operation. Reducing press speed may improve die filling, but it will not necessarily correct a blend whose API concentration has already become uneven.

Examine Size Fractions Before Removing Fines

Different granule fractions may contain different API concentrations. Removing fines or oversized granules can therefore change the remaining batch composition rather than simply improving flow.

Review the formulation, particle size distribution, and assays of relevant fractions before proposing screening changes. If milling is implicated, evaluate screen size, operating speed, feed conditions, and granule breakage using suitable pharmaceutical milling equipment.

Ingredient Redistribution During Granulation and Drying

Check Binder Distribution and Granulation Consistency

In wet granulation, differences in wetting and binder distribution can create granules with different structures or compositions. If part of the API dissolves in the granulation liquid, subsequent liquid movement can affect where the API is deposited during drying.

Compare binder addition records, mixing conditions, wet massing endpoints, and granule properties with acceptable batches. Investigate changes in liquid quantity, addition rate, and batch loading. Avoid changing granulation liquid or binder concentration without evaluating the effects on product quality.

For an established wet granulation route, assess whether the controls available on a 고전단 믹서 과립기 support the required binder delivery and mixing conditions. Equipment capability should be verified against the formulation through trials.

Distinguish Migration From Uneven Moisture Removal

During drying, dissolved ingredients may move with liquid toward evaporating surfaces. Migration can occur within individual granules or between regions of a stationary granule bed. Research comparing drying methods shows that migration behavior depends on the material system and drying conditions. [2]

An uneven moisture profile is a useful investigation signal, but moisture measurements alone do not demonstrate whether the API has redistributed. Where migration is suspected, combine moisture and temperature records with assay data from relevant locations or granule fractions.

Review bed loading, product temperature, airflow, drying time, and the established endpoint. Fluidization changes particle movement and heat and mass transfer, but it does not guarantee that migration is eliminated.

Account for Attrition and Selective Material Loss

Granule collisions and handling can generate fines. If the fines have a different API concentration from the bulk material, their capture in filters or removal by extraction can affect the composition of the recovered product.

Include material recovery and fines composition in the investigation when selective loss is plausible. Published fluid bed drying work links gas velocity with fines entrainment and product loss, supporting the need to consider airflow alongside the drying endpoint. [3]

When evaluating a 유동층 건조기, assess moisture control, attrition, filtration, and recovery using representative granules. Uniform final moisture is only one part of the evidence needed.

Verify Sampling and Analytical Reliability

A variable result can reflect actual product variation, sampling bias, sample preparation problems, or an analytical issue. Review the method and laboratory records alongside the manufacturing investigation.

Check whether the sampler disturbs or preferentially collects a particular fraction. Review sample size and location, extraction completeness, preparation steps, calculations, and instrument performance as appropriate. Preserve the original results and follow the site’s out-of-specification procedure; additional testing should investigate a justified hypothesis rather than continue until passing results are obtained. [4]

Confirm the Root Cause Through Process Evidence

Use a documented investigation to connect the suspected mechanism to the observed results. For example, if transfer segregation is suspected, compare appropriately collected material before transfer with material reaching the feeder and tablets produced across the run. An improvement limited to blender samples would not demonstrate that the downstream problem has been resolved.

Evaluate any proposed process change against other relevant attributes, including assay, dissolution, tablet strength, and material recovery. Reblending or regranulating a failed batch requires a documented quality assessment and should not be assumed to restore an acceptable product.

For equipment trials, provide the API loading range, material properties, batch sizes, transfer arrangement, process records, and failure pattern. Ask for a trial plan that measures the attribute under investigation. A blender, granulator, or dryer should be selected for its demonstrated suitability at the relevant process stage.

자주 묻는 질문

Can a batch pass assay and fail content uniformity

Yes. A composite assay describes the combined sample, while content uniformity evaluates individual dosage units. An acceptable average can coexist with excessive variation between tablets.

Why can blend samples pass while tablets fail

The blend samples may not represent the variation at the dosage unit scale. Alternatively, material may segregate after sampling during discharge, transfer, hopper storage, or feeding. Review both possibilities before extending the blending time.

Does a high shear granulator solve every uniformity problem

No. It may support an appropriate wet granulation process, but it cannot by itself resolve downstream segregation, an unsuitable sampling method, or selective material loss. Identify the mechanism and verify the proposed remedy with process data.

What information helps an equipment supplier assess the problem

Provide formulation characteristics, API loading, batch size, particle size and moisture data, process flow, sampling locations, and the timing of variable results. Include containment, cleaning, and utility requirements for any proposed equipment trial.
